- Nov 2011 - PresentPostdoctoral Research Scientist / University College DublinI am currently a postdoctoral researcher in the tumor biology group of Saint Vincents University Hospital. As part of the SFI SRC Molecular Therapeutics for Cancer Ireland, we are investigating novel targeted therapies for triple negative breast cancer. I am specifically working with various preclinical inhibitors provided from a variety of pharmaceutical companies that we are currently investigating for their anti-tumorigenic potential on triple-negative breast cancer cell lines. We also have a collaborative project with the University of Cambridge to investigate a novel monoclonal antibody developed by Dr. Gillian Murphy. Research outputs: the work was presented at an international conference and resulted in one peer-reviewed publication, plus one manuscript and one grant application currently pending evaluation.
- Aug 2010 - PresentPostdoctoral research assistant / University of Rochester Medical CenterI worked as a postdoctoral scientist in the group of Stephen Hammes at the University of Rochester Medical Center. Hammes's laboratory uses Xenopus laevis oocytes as a model for studying transcription-independent steroid-mediated molecular signaling. We were particularly interested in studying the control of protein translation at early stages of embryonic development through polyadenylation-binding proteins. This work has potential impact on cancer research, as work from other members of the lab has shown similar androgen-mediated translational control pathways are involved in prostate cancer.
- Aug 2006 - PresentPost-graduate Student / Research Assistant / Royal College of Surgeons in IrelandAchievements: demonstrated a novel role for rapid, estrogen-induced signalling in activating the eicosanoid signalling pathway to promote breast cancer cell proliferation. This work highlighted cPLA2α as a potential alternative target for therapeutic intervention in breast cancer, particularly in HER2-over-expressing mammary tumors. The project also involved a translational effort in collaboration with the department of surgery: we reported a novel direct correlation between high cPLA2 expression and poor prognosis in luminal cancer patients, suggesting a role for PLA2 in the early onset of resistance to endocrine therapy. Research outputs: this work was presented at two international conferences, selected for a peer-reviewed oral presentation at an irish national conference, and invited for oral presentation at a british national symposium, and resulted in two peer-reviewed papers and one review publication.
- Nov 2004 - PresentGraduate Student / Università degli Studi Roma TreI studied the signaling pathways activated by estrogen receptors (ERα and ERβ) at the plasma membrane. Specifically, we used a human colon cancer cell line (DLD-1), which only express the ERβ isoform, to study incorporation of radiolabeled palmitate in ERβ and to assess the ability of the receptor to associate with membrane proteins. We discovered that palmitoyl-acyltransferase , the enzyme responsible for ERα palmitoylation, can also promote ERβ palmitoylation with a different, less rapid kinetic. This modification is responsible for ERβ association with the plasma membrane and for the p38/MAPK activation, leading to induction of caspase-dependant cell death. In accordance with our results, a down-regulation of ERβ was recently linked with carcinogenesis in colon, as well as other tissues such as breast, ovary and prostate. Research outputs: this work produced two peer-reviewed papers and one review publication.
